[Dmt1]DALDA
Also known as [Dmt¹]DALDA; H-Dmt-D-Arg-Phe-Lys-NH₂
A dimethyltyrosine-substituted DALDA analogue for studying μ-opioid receptor recognition and peptide structure–activity relationships.
- Length
- 4 amino acids
- Half-life
- Not well characterised
- Mechanism
- [Dmt1]DALDA is a μ-opioid receptor-preferring agonist that activates Gi/o-coupled signalling. Replacing the N-terminal tyrosine of DALDA with 2′,6′-dimethyltyrosine modifies aromatic receptor interactions and can increase affinity and functional potency in experimental assays.
- Studied in
- Studies in receptor-expressing cells, isolated tissues and rodent models examine opioid signalling, peptide distribution and the effects of N-terminal dimethyltyrosine substitution.
About [Dmt1]DALDA
[Dmt1]DALDA has the sequence Dmt–D-Arg–Phe–Lys–NH₂, where Dmt denotes 2′,6′-dimethyltyrosine at the N terminus. It was developed through structure–activity investigations of DALDA, itself a shortened, modified analogue of the dermorphin opioid pharmacophore. The two methyl groups are attached to the aromatic ring of the first residue, rather than to the peptide backbone. Its free N terminus, D-arginine and C-terminal amide preserve the principal architectural features of the parent tetrapeptide.
The Dmt substitution changes the steric and hydrophobic environment around the phenolic opioid recognition group. Comparative assays commonly show stronger μ-opioid receptor binding and greater functional potency than DALDA, although the magnitude depends on experimental conditions. Research also examines membrane interactions and peptide stability, which should be distinguished from receptor activation itself. Despite its aromatic modification, the molecule remains strongly cationic, making buffer composition, matrix binding and analytical recovery relevant to interpreting laboratory measurements.
Laboratory notes
- Confirm that Dmt is 2′,6′-dimethyltyrosine with the specified stereochemistry; similarly abbreviated modified residues are not interchangeable.
- Compare with DALDA using the same receptor system, incubation period and functional endpoint.
- Check soluble peptide recovery and nonspecific binding when establishing low-concentration assay conditions.

Questions about [Dmt1]DALDA
What does Dmt1 mean?
It identifies replacement of the first residue with 2′,6′-dimethyltyrosine; the superscript or numeral specifies position one.
Is [Dmt1]DALDA identical to DALDA?
No. The added aromatic methyl groups create a distinct compound with different receptor-binding and functional properties.
Can binding affinity alone establish agonist potency?
No. Binding assays measure receptor association, whereas functional assays measure signalling. Receptor abundance and signal amplification can alter the relationship between them.
Is [Dmt1]DALDA for human use?
No. It is supplied strictly for in-vitro laboratory research and is not for human or animal consumption.
