Hormone · Amidated RFamide neuropeptide; GPR10 agonist
Prolactin-releasing peptide-20 (PrRP-20)
Also known as PrRP-20; PrRP20
An amidated 20-residue RFamide peptide used to investigate GPR10 signalling and receptor-dependent neuroendocrine responses.
- Length
- 20 amino acids
- Half-life
- Not well characterised
- Mechanism
- PrRP-20 activates GPR10, also called PRLHR, and can engage calcium mobilisation and downstream kinase signalling in responsive cells. It also shows activity at neuropeptide FF receptor 2 in experimental systems, making receptor-expression controls important for target attribution.
- Studied in
- Recombinant receptor assays, pituitary cell preparations and rodent brainstem–hypothalamic models investigate PrRP-20 receptor pharmacology and neuroendocrine signalling.
About Prolactin-releasing peptide-20 (PrRP-20)
PrRP-20 is the shorter of two principal prolactin-releasing peptide forms, comprising the C-terminal 20 residues of PrRP-31. Both arise from the PRLH-encoded precursor and terminate in an amidated arginine–phenylalanine motif characteristic of RFamide peptides. PrRP was identified in 1998 through work seeking an endogenous ligand for the orphan receptor GPR10. Its name reflects early pituitary secretion assays, rather than a complete description of its physiological roles or anatomical distribution.
GPR10, encoded by PRLHR, is the principal receptor associated with PrRP signalling. Recombinant systems commonly examine intracellular calcium and kinase responses, while neural preparations explore signalling in brainstem and hypothalamic networks. PrRP-20 can also interact functionally with neuropeptide FF receptor 2, complicating interpretation in cells expressing multiple RFamide receptors. The C-terminal amide is an important structural feature, and non-amidated material should not be assumed to reproduce the pharmacology of the native amidated peptide.
Laboratory notes
- Confirm C-terminal amidation in the sequence specification and analytical certificate; a free-acid analogue is a distinct reagent.
- Profile GPR10 and neuropeptide FF receptor expression before attributing calcium or kinase responses to a single target.
- For pituitary secretion assays, include cell-viability controls and verify immunoassay specificity in peptide-containing samples.

Questions about Prolactin-releasing peptide-20 (PrRP-20)
What is the relationship between PrRP-20 and PrRP-31?
PrRP-20 corresponds to the C-terminal 20-residue segment of PrRP-31. Both native forms share the amidated C-terminal RFamide motif.
Are GPR10 and PRLHR different receptors?
No. GPR10 is a commonly used receptor name, while PRLHR is its gene symbol and is also used as a receptor designation.
Is PrRP-20 selective enough to identify GPR10 activity on its own?
Not in every assay context. Activity at neuropeptide FF receptor 2 means that receptor-expression profiling and appropriate genetic or pharmacological controls are needed.
Is Prolactin-releasing peptide-20 (PrRP-20) for human use?
No. It is supplied strictly for in-vitro laboratory research and is not for human or animal consumption.
