Big Dynorphin

Also known as Dynorphin 32

A 32-amino acid peptide containing both Dynorphin A and B, Big Dynorphin is a non-selective opioid receptor agonist used in neurological research.

Length
32 amino acids
Half-life
Not well characterised, but longer than smaller dynorphin fragments.
Mechanism
This peptide contains the sequences of both Dynorphin A and Dynorphin B and acts as an agonist at kappa-, mu-, and delta-opioid receptors.
Studied in
It is studied in neuroscience to understand the processing of prodynorphin and the functional consequences of a multi-receptor opioid ligand.

About Big Dynorphin

Big Dynorphin is a 32-amino acid polypeptide that is an intermediate processing product of the prodynorphin precursor. Its structure uniquely contains the full sequence of Dynorphin A (1-17) at its N-terminus, followed by a Lys-Arg processing site, and the sequence of Dynorphin B (1-13) at its C-terminus.

Unlike its smaller cleavage products that show high selectivity for the kappa-opioid receptor, Big Dynorphin exhibits broader activity. It functions as an agonist at kappa, mu, and delta opioid receptors. This lack of selectivity makes it an interesting tool for studying integrated opioid system responses in various experimental setups.

Laboratory notes

  • Due to its length and charge, solubility may vary; test dissolution in small volumes of appropriate buffers.
  • Can be proteolytically cleaved into Dynorphin A and Dynorphin B, a factor to consider in experimental design.
  • Store lyophilised peptide at -20°C or colder for long-term stability.

Questions about Big Dynorphin

What is unique about Big Dynorphin's structure?

It is a larger peptide that contains the sequences of two other active opioid peptides, Dynorphin A and Dynorphin B.

Is Big Dynorphin a selective receptor agonist?

No, it is considered a non-selective agonist, with activity at kappa, mu, and delta opioid receptors.

What is the biological source of Big Dynorphin?

It is an intermediate product formed during the proteolytic processing of the prodynorphin protein in the central nervous system.

Is Big Dynorphin for human use?

No. It is supplied strictly for in-vitro laboratory research and is not for human or animal consumption.
