PYY(3-36)
Also known as Peptide YY (3-36)
PYY(3-36) is a peptide hormone fragment that acts on NPY2 receptors, used in laboratory models to study appetite regulation and energy balance.
- Length
- 34 amino acids
- Half-life
- Approx. 8-10 minutes
- Mechanism
- Functions as a high-affinity agonist for the neuropeptide Y receptor Y2 (NPY2R), primarily in the arcuate nucleus of the hypothalamus.
- Studied in
- Investigated in metabolic and neuroscience research for its role in the regulation of appetite, energy homeostasis, and gut motility.
About PYY(3-36)
PYY(3-36) is the major circulating form of Peptide YY, a 36-amino acid hormone secreted by L-cells of the distal gastrointestinal tract. The full-length PYY(1-36) is rapidly cleaved by the enzyme dipeptidyl peptidase-IV (DPP-IV) to form the PYY(3-36) fragment. This truncated form exhibits a distinct receptor binding profile from its precursor.
While PYY(1-36) binds to multiple NPY receptor subtypes, PYY(3-36) shows high selectivity for the NPY2 autoreceptor. This specificity makes it a valuable molecular probe in laboratory research to isolate and study NPY2R-mediated pathways involved in satiety signalling and energy expenditure. Its effects are often studied in ex vivo hypothalamic tissue and in animal models of metabolic regulation.
Laboratory notes
- PYY(3-36) is sensitive to proteolysis; use of protease inhibitors may be recommended for certain in vitro applications.
- For accurate results, ensure experimental models distinguish between the effects of PYY(3-36) and the full-length PYY(1-36).
- Lyophilised peptide should be stored at -20°C. Reconstitute with sterile, neutral-pH buffer just prior to use.

Questions about PYY(3-36)
What is the origin of PYY(3-36)?
It is an endogenously produced peptide, formed by the enzymatic cleavage of two N-terminal amino acids from the full-length Peptide YY (1-36).
Which receptor does PYY(3-36) primarily target?
It is a selective agonist for the presynaptic neuropeptide Y receptor subtype 2 (NPY2R).
Where is the primary site of action for PYY(3-36) in appetite research?
Its effects on satiety are largely mediated by its action on NPY2 receptors on NPY/AgRP neurons within the arcuate nucleus of the hypothalamus.
Is PYY(3-36) for human use?
No. It is supplied strictly for in-vitro laboratory research and is not for human or animal consumption.
